Kit Components

• Alphanaphtol Solution: 30 ml.
• Formalin/Ethanol Solution: 125 ml.
• Mayer Haematoxylin: 125 ml.
• Hydrogen Peroxide: 10 ml.
• Pyronin, 0,2% in aqueous solution: 125 ml.

Principle
Peroxidase granulations form an insoluble bright red compound with pyronin in presence of Alphanaphtol and Hydrogen Peroxide. The nuclei of leukocytes stain blue with Mayer Haematoxylin. This kit is benzidine-free. Benzidine is replaced by Alphanaphtol-Pyronin.

• Mayer Haematoxylin: 1 x 100 ml.
• Acid Buffer and Potassium Ferrocyanide: 10 x 5 ml.

Principle
In an acid environment, ferric ions react with potassium ferrocyanide what leads to a precipitate called Ferric Ferrocyanide (Prussian blue). It points out the presence of a pathological pigment: Haemosiderin.
